Edits not appearing

I will admit I am a total rookie with LRT, I have to refer to the instuctions on LRT.com every time. This confuses me though, When I edit the key frames, LR creates tif files that do not, appear as themselves, but instead a "second version" of the photo, and they don't show up is the catalog without doing other things, and therefore do not carry over the the rest of the sequence, what do I need to do to make this happen?

Sorry, I don't really understant.
When you edit the keyframes in Lightroom, no Tif files will be created. This will happen on a metadata basis only. You only "save" by invoking "Metadata/Save Metadata to files". BTW: Do not use Photoshop or any other external tool to edit the Raw files, this won't work!
Only in the very last step, when you export from Lightroom, you will be using the LRTExport plugin and this will create a JPG sequence (or Tiff, if you have the pro version). This will then be rendered to a video by LRTimelapse.
Please make sure to follow the "Visual Workflow" step by step as it's outlined here: https://lrtimelapse.com/workflow/

Question, do we have to edit in the Lr library, or can we edit with a saved custom preset in the develop module?

Of course you should edit in develop module. But I'd advise against using premade presets where you don't know what they do. This might lead to unexpected results. For example if you use different presets on different keyframes.
Background is that most of those presets use point curves (which are not supported for animation) or gradients (where you have to use the predefined gradients from the lrtimelapse initialization.
